PE-22-28 is an optimized analog of spadin that blocks the TREK-1 potassium channel, the same target some new antidepressants aim at. In rodents it produced antidepressant-like effects plus new hippocampal neuron and synapse growth in about four days, where SSRIs take weeks. It is entirely preclinical: no completed human trials, no human safety data.
- Blocks the TREK-1 potassium channel as an optimized analog of spadin.PubMed: PE-22-28
- In rodents it produced rapid antidepressant-like effects with increased hippocampal neurogenesis and synaptogenesis within about four days.PubMed: PE-22-28
- It is longer-acting than spadin, roughly 23 hours against about 7.PubMed: PE-22-28
- Spadin lacks SSRI-type side effects in animals, so the class looks theoretically favorable, though that is unverified. The Complete Peptide Guide p.79
